Diabetic nephropathy (DN) is a serious complication of diabetes that leads to end stage kidney disease (ESKD). Changes in the glomerular basement membrane (GBM) and the podocyte’s cytoskeleton are some of the earliest changes in the course of DN and are diagnostic for DN. Using super-resolution microscopy, STORM, I am studying the changes in the extracellular matrix composition and organization that could explain the increase in GBM thickness that is a hallmark change in DN. Also, I am studying the changes in key components of the podocyte actin cytoskeleton that could explain podocyte foot process effacement, the podocyte’s typical response to injury in DN and in other proteinuric kidney diseases.
